Serotonin is a neurotransmitter or a chemical in the brain that gives us feelings of pleasure and wellbeing. When serotonin is releases from cells in the brain or neurons it sends out signals to be happy and content. The “grouchy” gene is a genetic variant of the SLC6A4 gene, which provides instructions to make the serotonin transporter.

Individuals with the “grouchy” gene produce lower levels of the transporter, which affects serotonin levels in the brain. These changes result in an increased likelihood of mood disorders such as anxiety and depression.

Understanding the “Grouchy” Gene

The SLC64A gene, located on chromosome 17, determines the length of the 5-HTTLPR region. Inheriting one or two copies of the short (“grouchy”) form of this gene is associated with an increased risk of depression and unhappiness, while two copies of the long form decrease the risk of these conditions. Heterozygotes, carrying one copy of each form, also experience an impact on their level of happiness.

Selective serotonin reuptake inhibitors (SSRIs), such as Prozac, Zoloft, and Paxil, are commonly prescribed antidepressants. However, individuals with the “grouchy” gene variant may show reduced responsiveness to SSRIs due to lower serotonin transporter protein levels. This affects the efficacy of these medications, and alternative antidepressant options may be recommended.
